Since there is no more first step relating to a particular journey, we must not use “a” before “first step”. Having said that I would like to state that the more idiomatic way of the same sentence is “A journey of thousand miles starts [or begins] with a single step”. But it is not the perfect translation for the given Hindi sentence.
